Do I need to keep my computer switched on to scan it?

For all you computer experts, before you ask, yes I really AM a novice.
I'm confused - the MSE states its automatic so why is it not scanning automatically?
My setting is to scan Sunday night at 2am, so my big question is:

Do I have to leave my computer switched ON all night?

If so where does the "automatic" come into it? Do I have to remember every Sunday night to leave my computer on???
