Suspicious activity on my computer and failure to start Windows Defender

I cannot start Windows Defender because when launched a dialog box appears stating error: 0x800106ba was encountered. Further, a strange folder has appeared in my C drive which spontaneously disappeared a little while ago without any action by me.

The date and time has changed on my desktop, and I have lost the 'Show Desktop' icon from the taskbar of my Vista OS all within the same time period. I have run Microsoft Malicious Software Removal Tool, and performed multiple virus scans with AVG and Security Essentials, yet nothing has been detected. Any help would be greatly appreciated.
